What is the most appropriate and respectful way to identify an area for prayer in a workplace?   There is a colleague at work who is observing Ramadan and I would like to create a respectful do no disturbed sign so that people will not walk in on her during prayers

Now coming specifically to the issue you have raised: It is sufficient for this purpose to cordon off the specific area,  if it all possible, by setting up a barrier (permanent or temporary) so that people can be expected to respect the same, and in order to discourage them from walking in front of the worshippers while they are at it. If, however, setting up such barriers are problematic, then you may simply place prayer rugs to indicate the same.
